SEE ALSO...

ROD Antibody (43-K) is a mouse monoclonal IgG2a kappa light chain antibody that detects ROD protein of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Anti-ROD antibody (43-K) is available as the non-conjugated form. ROD, also known as KNTC1 (Kinetochore-associated protein 1), plays a critical role in the mitotic cell cycle checkpoint, where ROD (43-K) is essential for the assembly of MAD1-MAD2 and Dynein-Dynactin complexes at kinetochores. ROD is highly expressed in the testis and exhibits a dynamic localization pattern throughout the cell cycle, initially found in the nucleus and cytoplasm during interphase, and then translocating to kinetochores and spindle fibers during anaphase. ROD (43-K) functions to ensure accurate chromosome segregation, as ROD interacts with ZW10, forming a stable association with the kinetochore, which is crucial for spindle assembly. This interaction helps target the microtubule motor cytoplasmic Dynein to kinetochores, thereby facilitating the movement of chromosomes during cell division. ROD′s ability to regulate these processes underscores ROD′s importance in maintaining genomic stability and preventing aneuploidy, making ROD a significant target for research in cell biology and cancer studies.

I am using ROD (43-K): sc-81853 for immunofluorescence, what dilution of this antibody should I use?

Asked by: AbPolly
Thank you for your question. We recommend a starting dilution of 1:50. However, optimal antibody concentration should be determined by titration. Please find our full immunofluorescence protocol here: https://www.scbt.com/scbt/resources/protocols/immunofluorescence-cell-staining

Rated 5 out of 5 by from IF detectionROD antibody is a good antibody for IF detection for Hela cell ! I used it in a 1:200 dilution, but the antibody will most likely be useful in lower concentrations. The cells is PFA fixed and permabilized with 0.25 % saponin. Stained at RT for 1 hour.Hela cell can be used for positive control.
