孕酮受体抗体(AB-52)是一种IgG1κ小鼠单克隆PR抗体,可通过WB、IP、IF和IHC(P)检测小鼠、大鼠和人类来源的PR-A和PR-B。孕酮受体抗体(AB-52)既可作为偶联抗PR抗体形式,也可作为非偶联抗PR抗体形式。孕酮的作用由孕酮受体的两种功能不同的异构体PR-A和PR-B介导,它们由PR基因单拷贝内的不同雌激素可诱导启动子转录而来。PR-A中缺少PR-B的前164个氨基酸。与孕酮结合的PR-A和PR-B具有不同的转录激活特性。具体而言,PR-B在大多数细胞和启动子环境中起转录激活剂的作用,而PR-A在转录上不活跃,并作为类固醇激素受体转录活性的强配体依赖性反式显性抑制因子起作用。两种PR异构体中都存在映射到受体氨基末端的抑制域(ID)。有趣的是,ID仅在PR-A中具有功能活性,并且是PR-A对类固醇激素进行反式抑制所必需的,这表明PR-A和PR-B在细胞中可能具有不同的构象。
