Pax-8抗体(PAX8R1)是一种IgG1 κ小鼠单克隆Pax-8抗体,可通过蛋白质印迹、免疫沉淀、免疫荧光和流式细胞仪检测小鼠、大鼠和人源的Pax-8 A和Pax-8 B。Pax-8抗体(PAX8R1)为非偶联抗Pax-8抗体形式。Pax家族编码在胚胎发育过程中起作用的转录因子,并调节细胞的时间和位置依赖性分化。Pax-8在发育中和成年的甲状腺、发育中的分泌系统以及成年肾脏中较低水平表达。Pax-8与TTF-1和TTF-2形成复合物,通过调节钠碘转运体(NIS)、甲状腺过氧化物酶(TPO)、甲状腺球蛋白(TG)和促甲状腺素受体(TSHR)的表达来诱导甲状腺滤泡细胞分化和甲状腺激素的生物合成。用TGFβ1处理FRTL-5细胞可降低Pax-8 mRNA水平和Pax-8 DNA结合活性,从而抑制TG的表达和甲状腺细胞的形成。Pax-8基因发生常染色体显性突变的患者会出现甲状腺发育不良。Pax-8基因通过选择性剪接产生A到F六种异构体,它们在羧基末端区域有所不同。Pax-8异构体显示不同的DNA结合能力,并且在功能上被认为是有区别的。编码Pax-8的基因位于人类染色体2q12-q14上。
