p-Neu Antibody (14C3) is a mouse monoclonal IgG1 antibody that detects Neu in human samples through western blotting (WB). Neu, also known as v-ErbB-2 erythroblastic leukemia viral oncogene homolog 2 or HER2, plays a critical role in cell signaling pathways that regulate cell growth and differentiation, making Neu a significant factor in the development of various cancers, including breast, gastric, and esophageal cancers. Neu is primarily located in the cell membrane, where Neu interacts with other proteins to form signaling complexes essential for normal cellular function. Notably, Neu undergoes post-translational modifications, such as phosphorylation at specific tyrosine residues, which are crucial for activity and interactions. For instance, phosphorylation at Tyr 1248 facilitates Neu localization with the Mucin4/Sialomucin complex at the apical surfaces of ductal and alveolar cells, highlighting Neu′s importance in tissue architecture and function. Additionally, Neu phosphorylation at Tyr 1139 promotes association with GRB2 and GRB7, which are vital for downstream signaling pathways that can lead to tumorigenesis. Neu′s ability to undergo these modifications and interact with various signaling proteins underscores Neu′s role as a key player in oncogenic processes, making anti-Neu antibody (14C3) an invaluable tool for researchers studying cancer biology and therapeutic targets.
