I have a question. Please give me an answer. Can MMP-13 (C-3): sc-515284 monoclonal antibody be used in IHC with rat chondrocyte?

This antibody can distinguish whether MMP13 is active form or inactive? WB MMP13 band is not bad.

Thank you for your question. MMP-13 Antibody (C-3): sc-515284 is raised against amino acids 242-471 mapping at the C-terminus of MMP-13 of human origin. This epitope is conserved in both the inactive and active forms of MMP-13 so the antibody should be able to recognize both forms. However, we do not have data demonstrating this reactivity at this time.

Can MMP-13 (C-3): sc-515284 monoclonal antibody be used in IF or IHC with mouse tissue or cells? Will there be non-specific staining?

Thank you for your inquiry. The use of mouse monoclonal antibodies with mouse samples is very common and typically poses no problem. To eliminate any potential cross-reactivity of an anti-mouse secondary detection reagent, MMP-13 (C-3): sc-515284 is available in a variety of direct conjugations, such as HRP, PE, FITC and AlexaFluors.

Rated 4 out of 5 by from Good antibodyI tested this antibody for western blotting on whole lysate of different cell lines (30 µg). I used 1:500 dilution in 2.5% milk dissolved in TBS-0.1%Tween incubated over-night at 4°C. The antibody detects the specific band in BHT-101 cell line. Aspecific band at high molecular weight.

Using mouse brain tissue we were able to detect three discrete bands which appear to correspond with pro-MMP-13 (~60 kDa) and two cleavage fragments (~48 & ~25 kDa). We are very pleased with this antibody.
