MKP-2（MAP激酶磷酸酶2，双特异性蛋白磷酸酶4（DUSP4））是一种参与复杂的MAPKKK级联反应的磷酸酶。MKP-2属于蛋白酪氨酸磷酸酶家族（非受体类双特异性亚家族），包含一个硫氰酸酶结构域和一个酪氨酸蛋白磷酸酶结构域。MKP-2是一种双特异性蛋白磷酸酶，对MAPKs具有严格的底物特异性。它通过去磷酸化MAP激酶ERK 1和ERK 2上的Thr和Tyr残基来调节促有丝分裂信号转导。转录因子E2F-1负责介导细胞凋亡和抑制肿瘤发生，是MKP-2的转录调节因子。E2F-1与MKP-2启动子物理结合，可以反式激活MKP-2基因的启动子。具体而言，E2F-1与MKP-2启动子中的一个完美回文序列结合。MKP-2是E2F-1途径中一种重要的细胞死亡介体，可能导致癌症治疗新策略的开发。

What is the recommended fixation method for immunofluorescence staining with MKP-2 (48): sc-135991 monoclonal antibody?
