I have thought this antibody can equally recognise the phosphor- and non-phosphor-form of MEK-1/2, according to the statement "recommended for detection of the activation loop of MEK-1 and MEK-2 (independent of its phosphorylation status)". However, recently I did a growth factor stimulation experiment. I suddenly realised that MEK-1/2 Antibody (9G3) appears to have a strong preference for the p-MEK-1/2 over non-p-MEK-1/2, but is not totally clean with non-p-MEK-1/2. Thus, it would be problematic to use this antibody, especially when the phosphorylation level is low. So, please use this antibody with caution!

The epitope is in the activation loop, for sequential WB using a phosphor-MEK-1/2 and this Ab, whether the binding of one ab will interfere with the binding of the other is not unknown.
