    Overview

    Granzyme A (GZMA) is a tryptase serine protease stored in cytotoxic lymphocyte granules and released during immune synapse formation to promote target-cell damage and inflammatory signaling. In bovine systems, GZMA activity contributes to cell-mediated immunity by participating in proteolytic programs that can influence caspase-independent cell death, mitochondrial perturbation, and nucleic-acid damage responses. Beyond direct cytotoxicity, granzyme A has been linked to modulation of cytokine networks and innate immune pathways that shape leukocyte recruitment and antigen-driven responses. Altered granzyme-dependent effector functions are relevant to studies of immune dysregulation, chronic inflammation, and host–pathogen interactions in veterinary and comparative immunology.

    granzyme A CRISPR/Cas9 KO Plasmid (bovine) is a pool of plasmids designed for targeted disruption of the gene in bovine cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.

    The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish granzyme A protein expression.

    This CRISPR knockout system enables efficient generation of -deficient cell models for investigation of granzyme A signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
