ALDH7A1 encodes antiquitin, an NAD+-dependent aldehyde dehydrogenase that participates in lysine catabolism by oxidizing α-aminoadipic semialdehyde and related reactive aldehydes. Through detoxification of aldehyde intermediates, antiquitin contributes to cellular redox homeostasis and limits accumulation of electrophilic metabolites that can perturb metabolic and stress-response pathways. Loss-of-function variants are linked to pyridoxine-dependent epilepsy, reflecting disruption of lysine degradation and secondary effects on vitamin B6/PLP availability and neurotransmitter metabolism. ALDH7A1 is therefore a useful target for studying mitochondrial–cytosolic metabolic coupling, aldehyde stress biology, and inborn errors of metabolism in human cell models.
Antiquitin CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the ALDH7A1 gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the ALDH7A1 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the ALDH7A1 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish Antiquitin protein expression.
This CRISPR knockout system enables efficient generation of ALDH7A1-deficient cell models for investigation of Antiquitin signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
