Angiogenin (ANG), also referred to as ANG I, is a secreted member of the RNase A superfamily that regulates angiogenic signaling and stress-adaptive RNA metabolism. ANG can translocate to the nucleus and promote rRNA transcription, supporting ribosome biogenesis and proliferative capacity, and it also cleaves tRNAs to generate stress-induced fragments that modulate translation. Through these activities, ANG integrates extracellular cues with RNA processing programs that influence endothelial cell behavior, inflammation, and cellular survival under stress. Dysregulated ANG expression or activity has been associated with pathological vascular remodeling and neurodegenerative phenotypes, making it a useful node for mechanistic studies in angiogenesis and RNA stress pathways.
ANG I CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the ANG gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the ANG together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the ANG open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish ANG I protein expression.
This CRISPR knockout system enables efficient generation of ANG-deficient cell models for investigation of ANG I signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
