碱性磷酸酶（AP）是糖基-磷脂酰肌醇（GPI）锚定的、二聚体的、Zn2+金属化的糖蛋白，可催化磷酸单酯水解为无机磷酸和醇。存在至少四种不同但相关的碱性磷酸酶：肠（IAP）、胎盘（PLAP）、类胎盘（ALP-1或GCAP）和组织非特异性（TNAP）。前三种都位于2号染色体上，而组织非特异性形式位于1号染色体上。TNAP在肝、肾、骨、胃和结肠中广泛表达，因此被称为AP的组织非特异性形式。TNAP与血浆细胞膜糖蛋白-1结合，在骨矿化中发挥作用；然而，缺乏功能性TNAP形式的小鼠显示出正常的骨骼发育。这种酶已直接与一种被称为低磷酸酶症的疾病相关联，这是一种罕见的先天性疾病，其特点是骨骼矿化缺陷，包括骨骼缺陷。编码TNAP的人类基因定位于1p36.1-p34染色体。

To Santa Cruz, I have purchased the antibody for tissue non-specific Alkaline Phosphatase (TNAP) (D-3, sc-137213), and I can detect two bands between 70Kda and 100 Kda. I would like to know which one would be in fact the one corresponding to TNALP.

What is the recommended fixation method for immunofluorescence staining with TNAP (D-3): sc-137213 monoclonal antibody?
