GAPDH Antibody (G-9) is a mouse monoclonal IgG1 kappa light chain antibody raised against amino acids 1-335, representing the full-length glyceraldehyde-3-phosphate dehydrogenase (GAPDH) protein of human origin. Anti-GAPDH antibody (G-9) shows reactivity with mouse, rat, and human samples in various applications, including western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). GAPDH is a key enzyme in glycolysis, catalyzing the reversible oxidative phosphorylation of glyceraldehyde-3-phosphate in the presence of NAD+ and inorganic phosphate, a crucial step in energy metabolism. Beyond its metabolic role, GAPDH has multiple non-glycolytic functions, including involvement in transcriptional activation, RNA binding, DNA repair, and nuclear RNA export. Notably, GAPDH undergoes post-translational modifications such as phosphorylation and nitrosylation, which regulate its diverse functions and subcellular localization. These modifications are essential during cellular processes like apoptosis, where GAPDH translocates to the nucleus. GAPDH is also implicated in neurodegenerative diseases by interacting with proteins such as β-amyloid precursor and huntingtin. GAPDH monoclonal antibody (G-9) is available in both unconjugated and various conjugated forms, including horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and Alexa Fluor® conjugates.

We are using this antibody on mouse brain samples. We are getting a huge band at around 50kDa along with GAPDH. How can we use this antibody to get only the specific band of GAPDH?

why I get two clearly bands around ~55KD and ~40KD in my results,which one is specific to my antibody?thank you

Thank you for your question. The expected molecular weight for GAPDH is around 37-38kDa, so the band around 40kDa is your expected band.

How does GAPDH compare to Actin as a protein loading control for time course studies?

Thank you for your question. Both Actin and GAPDH are generally acceptable as loading controls for time course, RNAi and covalent mod studies. However, their molecular weights are slightly different; Actin (43kda), GAPDH (37kda). In addition, GAPDH is more relevant for non-differentiated lymphocytes, whereas Actin or GAPDH may be suitable for adherents.
