O anticorpo VDR (D-6) é um anticorpo monoclonal VDR de camundongo IgG2a κ (também designado anticorpo receptor de vitamina D ou anticorpo NR1I1) que detecta a proteína VDR de origem de camundongo, rato e humana por WB, IP, IF e IHC(P). O anticorpo VDR (D-6) está disponível na forma de anticorpo anti-VDR não conjugado, bem como em várias formas conjugadas de anticorpo anti-VDR, incluindo agarose, HRP, PE, FITC e vários conjugados Alexa Fluor®. O metabólito ativo da vitamina D modula a expressão de uma ampla variedade de genes de maneira específica ao desenvolvimento. Esse hormônio secosteroide pode regular para cima ou para baixo a expressão de genes envolvidos em uma série diversificada de respostas, como proliferação, diferenciação e homeostase do cálcio. A vitamina D3 1,25-(OH)2 exerce seus efeitos por meio da interação com o receptor de vitamina D (VDR), um membro da superfamília de receptores nucleares ativados por hormônios. Em seu estado ligado ao fixador, o VDR forma heterodímeros com o receptor de ácido retinóico 9-cis, RXR, e afeta a expressão gênica por meio da ligação de sequências específicas de DNA conhecidas como elementos de resposta a hormônios, ou HREs. Além de regular as respostas celulares mencionadas acima, a 1,25-(OH)2-vitamina D3 apresenta propriedades antiproliferativas em células de osteossarcoma, melanoma, carcinoma de cólon e carcinoma de mama.

Thank you for your question. Vitamin D Receptor/VDR Antibody (D-6): sc-13133 is recommended for detection of VDR of mouse, rat and human origin by WB, IP, IF and IHC(P), so it is suitable for immunocytochemistry.

does D6 vitamin d receptor antibody cross react with monkey (macaques) for immunohistochemistry

Thank you for your question. This antibody has not been tested on monkey samples in house, so it is not technically recommended or warrantied for that use, however, we do have publications in monkey (rhesus) samples.

Hello, I have been working with another VDR and I am not certain I am fully appreciating that VDR. Hence I am interested in the "Contact us to receive a FREE 10 µg sample of VDR (D-6): sc-13133." offer of getting 10 ug for free. Is it possible?

Can Santa Cruz provide VDR IHC images (performed using D-6 sc-13133 ab) with staining intensity (score) of 1 to 4 and % positive stained cells (score) with 1-4 (preferably in FFPE colorectal tumor tissue samples) ?

Thank you for the question. Please note that this product is sold for research or laboratory use only and it must not been used for medical diagnostics. All available IHC(P) images for this product will be published upon availability on our product website. We currently show staining of human parathyroid gland tissue showing nuclear and cytoplasmic staining of glandular cells, human skin tissue showing nuclear and cytoplasmic staining of keratinocytes, fibroblasts, Langerhans cells and melanocytes and human adrenal gland tissue showing nuclear staining of cortical cells. Kindly provided by The Swedish Human Protein Atlas (HPA) program.

Thank you very much for your inquiry. Please understand that we do not interpret customer data, since we do not have a detailed knowledge about your experiment and how the data was produced. Therefore our interpretation of the data could lead to a wrong direction without knowing all the relevant details. However, Western blotting is a technique that separates proteins based on size. In general, the smaller the protein the faster it migrates through the gel. However, migration is also affected by other factors, so the actual band size observed may differ from that predicted. Common factors include: 1. Post-translational modification - e.g. phosphorylation, glycosylation etc, which increases the size of the protein. 2. Post-translation cleavage - e.g. many proteins are synthesized as pro-proteins and then cleaved to give the active form, e.g. pro-caspases. 3. Splice variants - alternative splicing may create different sized proteins produced from the same gene. 4. Relative charge - the composition of amino acids (charged vs non-charged). 5. Multimers - e.g. dimerisation of a protein. This is usually prevented in reducing conditions, although strong interactions can result in the appearance of higher bands.
