SARA Antibody (F-9) is a mouse monoclonal IgG1 antibody that detects SARA in human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). SARA, also known as NSP, MADHIP, SMADIP, or ZFYVE9, is a double zinc finger protein characterized by its FYVE domain, which plays a crucial role in the recruitment of Smad proteins to the TGF-beta receptor, ensuring proper subcellular localization of the activated receptor-bound complex. The FYVE domain directs SARA to early endosomal compartments, where SARA interacts with TGF-beta receptors and Smads, facilitating the assembly of signaling complexes essential for TGF-beta signaling pathways. Notably, SARA′s interaction with the promyelocytic leukemia (PML) tumor suppressor enhances the association and accumulation of SARA and the TGF-beta receptor within early endosomes, thereby promoting effective signaling. Additionally, SARA is instrumental in recruiting protein phosphatase 1 catalytic subunit (PP1c) to the Smad7-GADD34 complex, which is vital for the dephosphorylation of the TGF-beta receptor by Smad7, a key mechanism for regulating negative feedback in TGF-beta signaling. anti-SARA antibody (F-9) is an invaluable tool for researchers studying TGF-beta signaling and its implications in various biological processes and diseases.
