PGRMC1 Antibody (C-4) is a mouse monoclonal IgM antibody that detects PGRMC1 of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). Anti-PGRMC1 antibody (C-4) is available as the non-conjugated format. PGRMC1, also known as progesterone receptor membrane component 1 or MPR, is a 195 amino acid single-pass membrane protein primarily localized to the endoplasmic reticulum and microsomes, where PGRMC1 plays a critical role in cellular signaling. PGRMC1 contains a cytochrome b5 heme-binding domain, essential for function as a receptor for progesterone, a steroid hormone vital for embryonic development and female reproductive health. PGRMC1 is expressed in various tissues, with highest levels found in the kidney and liver, highlighting PGRMC1′s importance in metabolic processes and hormonal regulation. The gene encoding PGRMC1 is located on human chromosome X, significant not only for its role in sex determination but also for association with various genetic conditions when abnormalities occur. Understanding PGRMC1 interactions with other proteins can provide insights into regulatory mechanisms and potential implications in reproductive health and disease.

Hi. We have stained kidney samples with PGRMC1 antibody (sc-393015, Lot #2217) but the results is different from the brochure (gromerulus: negative, tubules: positive). Is the lot wrong? How can I go ahead with the staining?

Rated 5 out of 5 by from Excellent validated antibodyThe SCBT PGRMC1 antibody (Sc393015) detects the native PGRMC1 protein at ~22 kDa and several post translational modified forms at ~65 and ~110 kDa molecular weight equivalent bands in HEK293 total cell extracts (see the attached immunoblot picture). The higher molecular weight bands are possibly sumoylated forms (see Peluso et al. 2012, PMID:22719051) and trafficked to different cell organelles. The antibody is also equally good in immunofluorescence study (see the attached image). The antibody detected ER and nuclear localized PGRMC1 which has been previously reported by Human Protein Atlas PGRMC1 validated Sigma antibody HPA002877. Interestingly this SCBT antibody is comparable (Immunoblot and immunofluorescence) to the Protein atlas validated antibody Sigma, but the price is half of the Sigma antibody. So, I would highly recommend this as a cost effective and efficient antibody for your research. Go ahead and use this antibody for your research and make great discoveries. It’s an important protein.

Rated 5 out of 5 by from western blot detectionThis antibody was used for the western blot detection of mouse PGRMC1 in primary lymphocytes. These cells were grown in RPMI with 10% FBS. This antibody is very specific and no background was seen in whole cell lysates.

Rated 5 out of 5 by from The antibody is very niceThis antibody been used for analysis of PGRMC expressin in human liver cell line,it gave the strong target band at 28kda when use the condition 1:400 for incubated 1hour at room temperature..Since it is IgM antibody.I recommend use the anti mouse IgM secondary.
