Ifna4 encodes interferon-α4 (IFN-α4), a type I interferon produced in response to viral nucleic acids and other innate immune triggers. IFN-α4 engages the IFNAR receptor complex to activate JAK1/TYK2-dependent phosphorylation of STAT1/STAT2 and formation of the ISGF3 transcriptional complex, inducing interferon-stimulated genes that restrict pathogen replication and modulate antigen presentation. This signaling shapes crosstalk with NF-κB and IRF pathways, influencing inflammatory gene programs, myeloid activation, and adaptive immune priming. Dysregulated type I interferon activity is relevant to chronic inflammation and autoimmunity research, as well as host–pathogen interaction models where interferon tone affects disease susceptibility and tissue pathology.
IFN-α4 CRISPR/Cas9 KO Plasmid (m) is a pool of plasmids designed for targeted disruption of the Ifna4 gene in mouse cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the Ifna4 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the Ifna4 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish IFN-α4 protein expression.
This CRISPR knockout system enables efficient generation of Ifna4-deficient cell models for investigation of IFN-α4 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
