O anticorpo anti-E-caderina (67A4) é um anticorpo monoclonal de cadeia leve IgG1 kappa de camundongo que detecta a E-caderina humana por western blotting (WB), imunoprecipitação (IP), imunofluorescência (IF) e citometria de fluxo (FCM). O anticorpo anti-E-caderina (67A4) está disponível em formas não conjugadas e várias formas conjugadas, incluindo agarose, peroxidase de rábano (HRP), ficoeritrina (PE), isotiocianato de fluoresceína (FITC) e vários conjugados Alexa Fluor®. A E-caderina, um membro da família das caderinas, desempenha um papel crucial na adesão célula-célula, essencial para manter a integridade dos tecidos e facilitar os processos morfogenéticos durante o desenvolvimento. A E-caderina localiza-se principalmente na membrana celular, formando junções aderentes que ligam células adjacentes e contribuem para a estabilidade estrutural dos tecidos. Esta localização na membrana permite à E-caderina mediar uma adesão intercelular extremamente forte, necessária para a formação da camada epitelial e para a prevenção de comportamentos invasivos nas células cancerosas. A interação da E-caderina com proteínas citoplasmáticas, como a β-catenina, é fundamental para as vias de sinalização que regulam a proliferação e a diferenciação celular, realçando a sua importância tanto na fisiologia normal como nos estados de doença.

Rated 1 out of 5 by from Anticorpo non funzionantePrimary antibodies E-Cadherin (67A4) sc-21791 was used diluted 1:500 according to the manufacturer’s recommendations (for Western Blotting dilution range 1:100-1:1000), in Milk 5%. Membrane of PVDF has been incubated overnight at 4°C and then it has been probed for 1h at RT with Santa Cruz secondary antibodies diluted 1:5000 in Milk 1%. We have tested this antibody on two different sets of samples: in six cancer cell lines and in human activated PBMCs. Moreover, even if we tested the antibody at higher concentration, we only obtained a higher background but not an increase in the specific protein signal; therefore we show only the blots obtained after the incubation with antibody prepared as described above. In conclusion: • E-Cadherin did not work at all, neither after a long exposure. We do not have tools to verify the specificity for this antibody too. All the membranes have been probed with a normalizer and each gel has been stained with Coomassie Brillant Blue, shown on the right side of each panel, in order to demonstrate that the samples ran well and that the proteins are present on the SDS-PAGE.
