Adrenomedullin (ADM) Ativadores

Adrenomedullin (ADM) is a multifunctional peptide hormone that plays a pivotal role in several physiological processes, including vasodilation, cell growth, and immune response modulation. It was initially discovered in human pheochromocytoma tissue and is widely expressed in various organs such as the lungs, kidneys, and cardiovascular system. The primary ADM signaling pathway involves its binding to a G protein-coupled receptor (GPCR) complex consisting of the calcitonin receptor-like receptor (CLR) and receptor activity-modifying proteins (RAMPs). This receptor binding activates adenylate cyclase, leading to an increase in cyclic AMP (cAMP) levels within the cell. Elevated cAMP activates protein kinase A (PKA), which then phosphorylates various downstream targets to mediate the diverse physiological effects of ADM. Besides the canonical CLR/RAMP receptor pathway, ADM also interacts with other receptor types, such as L1 cell adhesion molecule, to engage in more complex signaling networks.

Adrenomedullin (ADM) Activators constitute a chemical class of molecules designed to modulate the activity or expression of adrenomedullin. These compounds may operate through various mechanisms such as enhancing the binding affinity of ADM to its receptors, inhibiting pathways that oppose ADM action, or epigenetically modulating the expression of ADM. While some ADM activators are small molecules that can easily penetrate cellular membranes, others may be biologics or peptide-based compounds that mimic ADM's structure and function. Furthermore, these activators might also function by increasing the availability of essential substrates or cofactors required for ADM synthesis or activity.
