Y-27632 dihydrochloride monohydrate is a highly potent, cell-permeable, selective Rock (Rho-associated coiled coil forming protein serine/threonine kinase) inhibitor. Ki= 140 nM for p160 ROCK. It also inhibits ROCK-II with equal potency where the inhibition is competitive with respect to ATP. Y-27632 is a versatile research tool known for its range of biological effects, particularly in the field of stem cell research. It is employed as a key component in 3D growth matrices and brain organoid differentiation media. Notably, Y-27632 plays a role in the maintenance and differentiation of stem cells. Its use has been instrumental in enabling the survival and differentiation of human embryonic stem cells into cortical and basal telencephalic progenitors, thereby facilitating the study of developmental processes and potential therapeutic applications.
