UCH-L1 Antibody (31A3) is a mouse monoclonal IgG1 antibody that detects UCH-L1 in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and immunohistochemistry. UCH-L1, or ubiquitin C-terminal hydrolase, plays a crucial role in the ubiquitin-proteasome pathway by hydrolyzing small C-terminal adducts of ubiquitin to release free ubiquitin monomers, which are essential for protein degradation and cellular regulation. UCH-L1 is predominantly expressed in neurons and cells of the diffuse neuroendocrine system, with significant localization in brain regions such as the substantia nigra, where UCH-L1 constitutes 1-2% of total soluble brain protein. UCH-L1′s presence in Lewy bodies highlights UCH-L1′s potential involvement in neurodegenerative diseases, particularly Parkinson′s disease, making UCH-L1 a valuable target for research. The gene encoding UCH-L1 is located on human chromosome 4p13, and UCH-L1′s structural similarity to UCH-L3, which is 54% identical and serves as a predominant thiol protease with high-affinity binding sites for ubiquitin, underscores UCH-L1′s importance in maintaining cellular homeostasis and implications in neurodegenerative disorders.
