S-(+)-Rolipram is a stereoisomer of the cyclic AMP-specific phosphodiesterase (PDE IV) inhibitor Rolipram (sc-3563). S-(+)-Rolipram is demonstrated to be ten times less potent at PDE IV inhibition than (R)-(−)-Rolipram (sc-201199), and was similarly described to block antigen-induced contracts of guinea pig colonic smooth muscle ten times less effectively than (R)-(−)-Rolipram. Racemic preparations of Rolipram are described to confer neuroprotection against excitotoxic insult, and to increase wakefulness in rats through increasing cAMP availability and increasing noradrenaline release through cAMP-mediated stimulation of tyrosine hydroxylase activity.
