RPGR (retinitis pigmentosa GTPase regulator) encodes a ciliary protein enriched at the connecting cilium of photoreceptors, where it supports intraflagellar transport and trafficking of membrane-associated cargos required for outer segment maintenance. RPGR interacts with cilia-associated complexes and contributes to microtubule-based transport processes that preserve cilium structure and signaling competence. Disruption of RPGR perturbs ciliary function and photoreceptor homeostasis, linking its loss of function to inherited retinal degeneration phenotypes such as X-linked retinitis pigmentosa. In biomedical research, RPGR is commonly studied in the context of ciliopathy mechanisms, photoreceptor cell biology, and genotype–phenotype relationships in retinal disease.
RPGR CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the RPGR gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the RPGR together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the RPGR open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish RPGR protein expression.
