Protein S Antibody (PS7) is a rat monoclonal IgG2a antibody that detects Protein S of human origin by western blotting (WB), immunoprecipitation (IP), and flow cytometry (FCM). Anti-Protein S antibody (PS7) is available as the non-conjugated form. Protein S (PROS) is a crucial vitamin K-dependent plasma protein that plays a significant role in blood coagulation regulation by acting as a cofactor for activated protein C (APC), thereby inhibiting clot formation and promoting early apoptotic cell clearance. PROS is primarily located in plasma, where PROS exists in a dynamic equilibrium between active and inactive forms; approximately 60-70% of circulating PROS is bound to C4b-binding protein (C4BP), which renders PROS inactive. This interaction influences complement activation regulation and immune response. Additionally, PROS exhibits APC-independent anticoagulant activity, directly inhibiting prothrombin activation through interactions with Factor Xa, Factor Va, and phospholipids. A deficiency in PROS, particularly of the autosomal dominant type, can lead to significantly reduced levels (15 to 37% of normal) and is associated with severe recurrent venous thrombosis, highlighting PROS importance in maintaining hemostatic balance.
