PKR CRISPR/Cas9 KO Plasmid (rabbit) | sc-437324 | 20 µg | $397.00 |
PKR (EIF2AK2) is an interferon-inducible serine/threonine kinase that senses double-stranded RNA and coordinates innate antiviral defenses. Upon activation, PKR phosphorylates eIF2α to suppress cap-dependent translation, shaping cellular stress responses and modulating NF-κB and IRF signaling programs. PKR activity influences apoptosis, inflammasome-related outputs, and crosstalk with MAPK pathways, linking pathogen sensing to broader gene-expression control. Dysregulated PKR signaling has been associated with chronic inflammation, viral susceptibility, and altered proteostasis relevant to neurodegeneration and metabolic stress models.
PKR CRISPR/Cas9 KO Plasmid (rabbit) is a pool of plasmids designed for targeted disruption of the gene in rabbit cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish PKR protein expression.
This CRISPR knockout system enables efficient generation of -deficient cell models for investigation of PKR signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
