Mouse Pax1 encodes the paired box transcription factor Pax-1, a nuclear DNA-binding protein that regulates lineage specification and patterning during embryogenesis, particularly within paraxial mesoderm and the developing vertebral column. Pax-1 integrates developmental signaling inputs to control transcriptional programs involved in chondrogenesis, sclerotome differentiation, and segmental organization of the axial skeleton. Altered Pax1 function has been linked to congenital vertebral and skeletal malformations and is used as a marker and regulator in studies of spine formation and musculoskeletal development. In cellular models, Pax-1 activity is investigated for its effects on transcriptional networks governing proliferation and differentiation in mesenchymal and cartilage progenitor contexts.
Pax-1 CRISPR/Cas9 KO Plasmid (m) is a pool of plasmids designed for targeted disruption of the Pax1 gene in mouse cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the Pax1 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the Pax1 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish Pax-1 protein expression.
This CRISPR knockout system enables efficient generation of Pax1-deficient cell models for investigation of Pax-1 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
