PACT Antibody (2830C1a) is a mouse monoclonal IgG1 antibody that detects PACT in human samples through applications such as western blotting (WB) and immunoprecipitation (IP). PACT, or PKR-associated protein X, plays a crucial role in the cellular response to stress by activating the protein kinase PKR, which subsequently leads to the phosphorylation of eIF2α, a key regulator of protein synthesis. This process is vital for controlling cellular translation and promoting apoptosis under unfavorable conditions, such as serum starvation or oxidative stress. Notably, PACT can directly bind to double-stranded RNA, although PKR activation and resulting eIF2α phosphorylation occurs primarily in the absence of dsRNA. The phosphorylation of PACT at Serine 18 is particularly significant, as this modification enhances PKR activation following stress exposure. Additionally, PACT may form a heterodimer with eIF2α, utilizing its double-stranded RNA-binding motif (DRBM) domain for interaction, which underscores PACT′s importance in the regulation of cellular stress responses and apoptosis. PACT (2830C1a) antibody serves as an essential tool for researchers studying these critical pathways in human cells.

Thank you for your question. We recommend using 1–10 µl (i.e., 0.2–2 µg) of antibody with 1 ml of cell lysate. However, optimal antibody concentration should be determined by titration.
