p22-phox Antibody (44.1) is a mouse monoclonal IgG2a kappa light chain antibody that targets the p22-phox protein, also known as CYBA or cytochrome b-245 alpha chain, from human origin. anti-p22-phox antibody (44.1) recognizes specific epitopes at amino acids 29-33 and 182-188, enabling detection of p22-phox in mouse, rat, and human samples through western blotting (WB), immunoprecipitation (IP), and flow cytometry (FCM). p22-phox is an essential component of the NADPH oxidase complex, which plays a critical role in generating superoxide radicals in phagocytes, neuroepithelial cells, vascular smooth muscle cells, and endothelial cells. Together with gp91-phox (NOX2), p22-phox forms the membrane-bound flavocytochrome b558, necessary for the assembly and activation of the enzyme complex. During activation, cytosolic proteins p47-phox and p67-phox translocate to the plasma membrane to associate with p22-phox and gp91-phox, facilitating the production of reactive oxygen species that are vital for host defense and cellular signaling. Additionally, p22-phox acts as a surface oxygen sensor, initiating signaling pathways in response to hypoxic conditions. p22-phox monoclonal antibody (44.1) is available in multiple conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and various Alexa Fluor® conjugates, providing versatile options for diverse experimental applications. anti-p22-phox antibody (44.1) supports the investigation of critical pathways involved in cellular signaling and the body′s defense mechanisms against oxidative damage.

Hello, You state that this clone can recognize mouse p22. Do you have any example immunoblot or flow cytometry data for this antibody on mouse cells? Thanks, David

Thank you for your question. We have not yet tested this mouse monoclonal antibody, p22-phox (44.1): sc-130550, in mouse or rat samples. However, the antibody is expected to cross-react with mouse and rat based on sequence identity with the antigen used to develop the antibody. We do warranty the antibody for detection of p22-phox of mouse, rat or human origin.

Can p22-phox (44.1): sc-130550 monoclonal antibody be used in IF or IHC with mouse tissue or cells? Will there be non-specific staining?

Thank you for your inquiry. The use of mouse monoclonal antibodies with mouse samples is very common and typically poses no problem. To eliminate any potential cross-reactivity of an anti-mouse secondary detection reagent, p22-phox (44.1): sc-130550 is available in a variety of direct conjugations, such as HRP, PE, FITC and AlexaFluors.

Rated 4 out of 5 by from Good detection in protein extract from human bloodI tested the antibody at 1:1000 dilution in Everyblot solution (by Biorad), by overnight incubation at 4°C. In this conditions, I obtained a very good signal in samples from human blood, while I was not able to detect the signal in human immortalized fibroblasts.

Rated 5 out of 5 by from Perfect for FACS applicationWe bought the sc-130550 PE (i.e. the PE coupled clone 44.1) and tested it on both PBMCs from a normal donor or from a p22phox mutated patient. The signal is absolutely perfect and specific since only the control shows a nice and strong peak.
