Datasheets
  • p-p53 Antibody (B-12) is a mouse monoclonal IgG1 κ provided at 200 µg/ml
  • specific for an epitope mapping between amino acids Thr 155 Thr 155 of p53 of human origin
  • recommended for detection of Thr 155 phosphorylated p53 of human origin by WB, IP, IF and ELISA
  • m-IgG Fc BP-HRP and m-IgG1 BP-HRP are the preferred secondary detection reagents for p-p53 Antibody (B-12) for WB applications. These reagents are now offered in bundles with p-p53 Antibody (B-12) (see ordering information below).

    SEE ALSO...

p-p53 Antibody (B-12) is a mouse monoclonal IgG1 antibody that detects p53 in human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). anti-p53 antibody (B-12) recognizes phosphorylated forms of p53, a crucial tumor suppressor protein that plays a vital role in cellular stress responses by regulating the expression of genes involved in growth arrest and apoptosis. p53 monoclonal antibody (B-12) targets p53 primarily located in the nucleus, where p53 functions as a transcription factor to maintain genomic stability and prevent tumorigenesis. Nuclear translocation to the cytoplasm, often mediated by the negative regulator MDM2, highlights the importance of nuclear localization in controlling cell cycle progression and apoptosis. MDM2, an E3 ubiquitin ligase, can polyubiquitinate p53, leading to degradation, which underscores the delicate balance between p53 activity and cellular homeostasis. Interactions with proteins such as 14-3-3σ enhance p53′s stability and nuclear retention by recognizing phosphorylated serine residues, while the acetyltransferase p300 further activates p53 by acetylating lysine residues, enabling stronger DNA binding and transcriptional activity. Furthermore, phosphorylation at specific residues, such as Thr 155, can promote interactions with DNA repair proteins, including BRCA1 and Rad51, emphasizing p53′s role in genomic stability. These complex networks of interactions demonstrate how post-translational modifications like phosphorylation fine-tune p53′s activity and interactions, making p53 (B-12) monoclonal antibody an invaluable tool for studying the dynamics of p53 regulation in cancer research and therapeutic development.

Hi, 2 questions regarding sc-377564: In one of the two images, you see p-p53 at 89kDa! How can that be?? And one of the reviews yo are talking about good signals against p53. But the antibody is supposed to recognize p-p53. Can you clarifiy this?

    Asked by: Rebe
Thank you for your question. The first image shows data with human recombinant p53, which does not appear at the same molecular weight as wildtype p-p53. The antibody is only specific against phosphorylated p53 and will not recognize the non-phosphorylated protein.

What application is the blocking peptide sc-377564 P appropriate for?

    Asked by: Dr Ninau Qelp
Thank you for your question. The blocking peptide is intended for use as a negative control, by pre-adsorbing the mouse monoclonal antibody against the antigen. For full protocol details, please contact our Technical Services Department or view our online protocol here: https://www.scbt.com/scbt/resources/protocols/peptide-neutralization
