NLE1 Antibody (G-5) is a mouse monoclonal IgG1 kappa light chain antibody that detects NLE1 of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). NLE1 (G-5) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. NLE1, also known as notchless homolog 1, is a 485 amino acid WD40-repeat protein that plays a critical role in regulating the Notch signaling pathway, which is essential for proper cell communication during development. NLE1 is primarily located in the cytoplasm, where NLE1 interacts with the cytoplasmic domain of Notch receptors, thereby modulating their signaling activity. NLE1′s cellular location is important for influencing cell fate decisions and developmental processes; deletion of NLE1 during early embryonic development in mice leads to embryonic lethality, while later deletion activates a caspase-3-dependent apoptotic pathway, highlighting NLE1′s vital role in maintaining cellular integrity. In plants, NLE1 is crucial for normal growth and development, with under-expression during shoot proliferation resulting in delayed flowering and abnormal organ maturation. Additionally, NLE1 may be involved in the biogenesis of the 60S ribosomal subunit in yeast. NLE1′s structure, characterized by eight WD40 domains, allows for versatile interactions, and NLE1 produces one isoform through alternative splicing, further emphasizing NLE1′s functional significance in various biological contexts.

What is the recommended fixation method for immunofluorescence staining with NLE1 (G-5): sc-377142 monoclonal antibody?

Asked by: Germaine
Thank you for your inquiry. We recommend fixing cells for 5 minutes in -10°C methanol and allowing to air dry. The full protocol can be viewed here: https://www.scbt.com/scbt/resources/protocols/immunofluorescence-cell-staining
