SEE ALSO...

Mel-CAM Antibody (OJ79c MUC 18) is a mouse monoclonal IgG1 antibody that detects Mel-CAM in human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and flow cytometry (FCM). Mel-CAM, also known as MCAM, MUC18, or CD146, is a transmembrane glycoprotein encoded by the human Mel-CAM gene, which plays a crucial role in cell adhesion and is a member of the immunoglobulin superfamily. This protein consists of 603 amino acids, featuring a signal peptide, five immunoglobulin-like domains, a transmembrane region, and a short cytoplasmic tail, which are essential for its function. Mel-CAM is predominantly expressed in advanced primary and metastatic melanomas, as well as in neuroectodermal lineage cell lines, but is absent in normal melanocytes. Mel-CAM expression is found in approximately 80% of advanced primary human melanomas and is closely associated with the progression of metastatic disease. Mel-CAM activation triggers an outside-in signaling pathway involving protein tyrosine kinases such as Fyn, Focal Adhesion Kinase (FAK), and paxillin, which are vital for the dynamics of actin cytoskeleton rearrangement. This process is not only critical for tumor cell migration and invasion but also plays a significant role in maintaining thymic architecture and function, highlighting Mel-CAM′s importance in both cancer biology and immune system integrity.

How can Mel-CAM (OJ79c MUC 18): sc-53369 mouse monoclonal antibody be used for double or triple staining, if the other primary antibodies are also raised in mouse?

Asked by: Cweed
In this case, we recommend using a directly conjugated mouse monoclonal primary antibody. This antibody is available conjugated to phycoerythrin (sc-53369 PE) or fluoroscein (sc-53369 FITC).
