    Mel-18 Antibody (B-8) is a mouse monoclonal IgM antibody that detects Mel-18 in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Mel-18, a member of the Polycomb group gene family, plays a crucial role in the regulation of gene expression during development, particularly in the repression of homeotic loci, which is essential for proper embryonic development. Mel-18 (B-8) antibody targets a protein primarily located in the nucleus, where Mel-18 interacts with chromatin to modulate transcriptional activity. Nuclear localization of Mel-18 allows for direct engagement with DNA and associated regulatory elements, thereby influencing the expression of key genes involved in cell proliferation and survival, such as Bcl-2 and c-Myc. Furthermore, Mel-18′s ability to bind DNA through its RING-finger motif facilitates its role as a tumor suppressor, as Mel-18 negatively regulates cell cycle progression and prevents inappropriate entry into the S phase. In contrast, the oncogenic potential of its homolog Bmi-1 highlights the delicate balance of these proteins in cellular processes, making anti-Mel-18 antibody (B-8) a vital tool in understanding both normal development and cancer biology.

    Secondary antibody: could m-IgG kappa BP-HPR be used for detection, or an anti-IgM should be used instead ?

    Thank you for your question. We have not yet tested our Mouse IgG kappa Binding Protein (sc-516102) for detection of a primary mouse IgM by Western blotting. It may work, but we would recommend using an isotype-specific anti-mouse IgM for detection of this Mel-18 Antibody (B-8): sc-390868. Unfortunately, we do not offer this type of secondary currently, but we are developing one and hope to release it soon.
