KCTD5 CRISPR/Cas9 KO Plasmid (h) | sc-409961 | 20 µg | $397.00 |
KCTD5 encodes a potassium channel tetramerization domain–containing protein that functions as an adaptor in ubiquitin-dependent protein homeostasis, commonly acting with CUL3-based E3 ligase complexes to influence substrate ubiquitination and turnover. Through these interactions, KCTD5 can impact signaling network dynamics and cellular proteostasis, including processes linked to neuronal function and stress-responsive pathways. Altered regulation of KCTD family adaptors has been associated with dysregulated protein degradation and perturbed signaling outputs, which are recurrent features in cancer biology and neurobiological disorders. Consequently, KCTD5 is studied as a modulatory node connecting ubiquitination machinery to pathway control and cellular phenotype.
KCTD5 CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the KCTD5 gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the KCTD5 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the KCTD5 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish KCTD5 protein expression.
This CRISPR knockout system enables efficient generation of KCTD5-deficient cell models for investigation of KCTD5 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
