JAM-A Antibody (J10.4) is a mouse monoclonal IgG1 kappa light chain antibody raised against a recombinant JAM fusion protein of human origin, specifically reactive with JAM-A from mouse, rat, and human species as detected by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and flow cytometry (FCM). Junctional Adhesion Molecule A (JAM-A), also known as JAM-1 or F11 Receptor (F11R), is a member of the immunoglobulin superfamily prominently expressed at tight junctions of epithelial and endothelial cells. JAM-A interacts with various proteins such as ZO-1, AF-6, and PAR-3, which are PDZ domain-containing proteins crucial for tight junction assembly and maintenance. These interactions regulate cell polarity and paracellular permeability, essential for tissue homeostasis. Additionally, JAM-A interacts with integrins like LFA-1 (CD11a/CD18) on leukocytes, facilitating their transmigration across the endothelium during immune responses. JAM-A also associates with other tight junction proteins and signaling molecules, influencing cellular adhesion and motility. By studying these protein-protein interactions, researchers can gain insights into physiological processes such as inflammation, barrier function, and leukocyte extravasation. Anti-JAM-A antibody (J10.4) provides a specific and reliable tool for investigating JAM-A′s role in various biological contexts, making this reagent valuable for advancing our understanding of potential therapeutic targets.

Using this antibody (sc-53623), I get two bands in WB in tissues, but only one band in cell lysates. I want to know why

Can this antibody be used for staining mouse tissue?

Yes. We recommend using a directly conjugated antibody. JAM-A Antibody (J10.4): sc-53623 is available conjugated to HRP (sc-53623 HRP), 200 µg/ml, for WB, IHC(P) and ELISA; phycoerythrin (sc-53623 PE), fluorescein (sc-53623 FITC), Alexa Fluor® 488 (sc-53623 AF488) or Alexa Fluor® 647 (sc-53623 AF647), 200 µg/ml, for IF, IHC(P) and FCM.
