IRX3 CRISPR/Cas9 KO Plasmid (h) | sc-404632 | 20 µg | $397.00 |
IRX3 (Iroquois homeobox 3) encodes a homeobox transcription factor that helps establish transcriptional programs during embryonic patterning and organogenesis, with context-dependent roles in cell fate specification and differentiation. In human cells, IRX3 integrates with developmental signaling networks, including pathways linked to Wnt and BMP signaling, to shape lineage decisions and tissue-specific gene expression. Beyond development, altered IRX3 regulation has been associated with metabolic phenotypes and changes in adipocyte biology, and dysregulated IRX3 expression has been reported across multiple cancer contexts. These features make IRX3 a useful node for studying transcriptional control, differentiation state, and pathway-driven gene regulatory circuitry.
IRX3 CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the IRX3 gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the IRX3 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the IRX3 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish IRX3 protein expression.
This CRISPR knockout system enables efficient generation of IRX3-deficient cell models for investigation of IRX3 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
