Int-6 Antibody (A-1) is a mouse monoclonal IgG1 antibody that detects Int-6 in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Int-6, also known as eIF3e or eIF3-p46, plays a crucial role in regulating translation and protein degradation by interacting with several key complexes, including the eukaryotic translation initiation factor 3 (eIF3), the proteasome regulatory lid, and the constitutive photomorphogenesis 9 signalosome. The eIF3 complex is essential for the assembly of 40S ribosomal subunits on mRNA, which is vital for initiating protein synthesis. Notably, the Int-6 gene is a significant site of integration for the mouse mammary tumor virus (MMTV) in murine tumors, highlighting Int-6′s potential role in cancer biology. Furthermore, studies have shown that reducing Int-6 expression through RNA interference in HeLa cells can lead to defects in mitosis, including issues with spindle formation, chromosome segregation, and cytokinesis. These mitotic aberrations are associated with the inhibition of cyclin B-Cdk1 kinase activity, resulting from a prolonged inhibitory phosphorylated state of Cdk1. Int-6 (A-1) antibody is therefore an invaluable tool for researchers investigating the intricate mechanisms of translation regulation and its implications in cellular processes and disease states.
