His-Tag Antibody (HIS.H8) is a mouse monoclonal IgG2b antibody raised against a 6x His-tagged polypeptide, ideal for applications such as western blotting (WB), immunoprecipitation (IP), and immunofluorescence (IF). The His-tag consists of a sequence of six histidine residues added to either the N-terminus or C-terminus of a recombinant protein, and its structure is crucial for its function. The imidazole side chain of histidine has a strong affinity for metal ions like nickel and cobalt, allowing His-tagged proteins to bind efficiently to metal-chelating resins during immobilized metal affinity chromatography (IMAC). This structural property enables the purification of recombinant proteins in a single step while maintaining their native conformation and function. The small size of the His-tag minimizes interference with the protein′s structure and activity, making His-tag a preferred choice in protein expression studies. Understanding the structure and function of the His-tag is important for researchers involved in protein purification, detection, and analysis. why my protein doesn't show up from WB membrane, i have been used antibody anti his HIS.H8 (st.cruz) 1st antibody and mouse HRP conjugate (st cruz) as a 2nd antibody..what is wrong? is the 1st antibody not compatible with 2nd antibody? thank you
