    SEE ALSO...

Gfi-1 Antibody (15-8) is a mouse monoclonal IgG1 antibody that detects Growth factor independent 1 (Gfi-1) in human samples through various applications including western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). Gfi-1 plays a crucial role as a transcriptional repressor, specifically binding to the DNA consensus sequence TAAATCAC(A/T)GCA, which is vital for regulating gene expression involved in cell survival and differentiation. Gfi-1′s structure features six C2H2-type zinc finger motifs located at its carboxy-terminus, with zinc fingers 3, 4, and 5 being essential for DNA binding capability. A 20 amino acid SNAG domain in Gfi-1 facilitates transcriptional repression function, particularly inhibiting the pro-apoptotic gene Bax at both mRNA and protein levels, thereby preventing cell death. Beyond lymphoid system involvement, Gfi-1 is expressed in granulocytes and activated macrophages, highlighting importance in immune responses. Human Gfi-1 and related protein Gfi-1B genes, primarily expressed in bone marrow and spleen, are located on chromosomes 1p22 and 9q34.3, respectively, underscoring evolutionary conservation and functional significance in hematopoiesis and immune function.

Can sc-101053: Gfi-1 (15-8) monoclonal antibody be used to stain formalin-fixed, paraffin-embedded (FFPE) tissue sections?

    Asked by: Randy McDonald
Thank you for your question. Yes, sc-101053: Gfi-1 (15-8) is recommended for use in IHC with paraffin-embedded sections. We recommend performing antigen retrieval with sodium citrate buffer (pH 6) and heat. The full protocol can be found here: https://www.scbt.com/scbt/resources/protocols/immunoperoxidase-staining
