Mouse Cyc1 encodes cytochrome c1, an essential subunit of mitochondrial complex III (ubiquinol–cytochrome c reductase) that mediates electron transfer from ubiquinol to cytochrome c within the inner mitochondrial membrane. Through its role in the Q cycle, cytochrome c1 contributes to proton motive force generation that powers ATP synthesis and supports redox homeostasis. Disruption of complex III components can impair oxidative phosphorylation, elevate reactive oxygen species, and trigger secondary effects on apoptosis signaling and mitochondrial quality control pathways such as mitophagy. Cyc1 is therefore widely studied in models of mitochondrial dysfunction and metabolic stress that intersect with neuromuscular and neurodegenerative disease mechanisms.
cytochrome c1 CRISPR/Cas9 KO Plasmid (m) is a pool of plasmids designed for targeted disruption of the Cyc1 gene in mouse cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the Cyc1 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the Cyc1 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish cytochrome c1 protein expression.
This CRISPR knockout system enables efficient generation of Cyc1-deficient cell models for investigation of cytochrome c1 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
