Cypher Antibody (E-8) is a mouse monoclonal IgG2a kappa light chain antibody that detects Cypher protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Cypher (E-8) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Cypher protein, also known as LDB3 (LIM domain binding 3), ZASP (Z-band alternatively spliced PDZ-motif protein), ORACLE, and PDLIM6 (PDZ and LIM domain 6), is a 727 amino acid protein primarily expressed in skeletal muscle, with lower expression levels in brain, placenta, and heart. Cypher protein is localized to the perinuclear region of the cytoplasm and features three LIM zinc-binding domains, which are crucial for its role as an adaptor protein. These LIM domains facilitate the coupling of protein kinase C-mediated signaling in striated muscle to the cytoskeleton, thereby playing a vital role in maintaining muscle integrity and function. Defects in the gene encoding Cypher have been linked to several forms of dilated cardiomyopathy and myofibrillar myopathy, conditions characterized by impaired cardiac muscle structure and function. Six isoforms of Cypher exist due to alternative splicing, highlighting the complexity and regulatory potential of this protein in muscle biology.
