cSHMT Antibody (S-W6) is a mouse monoclonal IgG1 antibody that detects mammalian serine hydroxymethyltransferase (SHMT) in human samples through various applications including western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). Anti-SHMT antibody (S-W6) specifically targets the cytosolic form of SHMT, which plays a crucial role in cellular metabolism by catalyzing the reversible conversion of serine and tetrahydrofolate to glycine and methylenetetrahydrofolate. This enzymatic activity is vital for the synthesis of nucleotides, particularly thymidylate, which is essential for DNA replication and repair. Proper functioning of SHMT is critical for maintaining one-carbon metabolism, and any dysregulation can lead to metabolic disorders or contribute to the development of certain cancers. SHMT′s tetrameric structure, which is stabilized by specific residues such as Lys-71, Arg-80, and Asp-89, is essential for catalytic activity, as these residues influence intra-subunit ionic interactions. Additionally, active site residues, including Tyr-72, Asp-227, and His-356, interact with pyridoxal phosphate, further ensuring enzymatic functionality. Human SHMT gene is located on chromosome 17p11.2, and the mitochondrial variant is encoded by a gene on chromosome 12q13, highlighting SHMT′s importance in both cytosolic and mitochondrial metabolic pathways.

Thank you for your question. Yes, sc-100849: cSHMT (S-W6) is recommended for use in IHC with paraffin-embedded sections.
