Chd8 encodes chromodomain helicase DNA-binding protein 8 (CHD8), an ATP-dependent chromatin remodeler that regulates nucleosome positioning and transcriptional programs during development. CHD8 influences enhancer and promoter accessibility, linking chromatin architecture to cell-cycle control, neuronal differentiation, and Wnt/β-catenin–associated transcriptional outputs. In mouse systems, Chd8 is widely used to interrogate mechanisms of neurodevelopment and gene regulatory networks, as perturbation of CHD8 dosage has been connected to autism spectrum disorder–associated biology in genetic and functional studies. These properties make CHD8 a key node for studying chromatin-mediated control of lineage specification and transcriptional homeostasis.
CHD8 CRISPR/Cas9 KO Plasmid (m) is a pool of plasmids designed for targeted disruption of the Chd8 gene in mouse cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the Chd8 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the Chd8 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish CHD8 protein expression.
This CRISPR knockout system enables efficient generation of Chd8-deficient cell models for investigation of CHD8 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
