Caspase-3 inhibitor (Ac-AAVALLPAVLLALLAPDEVD-cho) is a cell-permeable inhibitor of caspase-3, caspase-6, caspase-7, caspase-8, and caspase-10. Caspase inhibitors play an important role in investigating biological processes. The caspase family of proteins function as key components of the apoptotic machinery and act to destroy specific target proteins which are critical to cellular longevity. The C-terminal sequence of this peptide (DEVD-CHO) is a highly specific, potent, and reversible inhibitor of caspase-3 that also strongly inhibits PARP cleavage in cultured human osteosarcoma cell extracts. Following use of the inhibitor, standard apoptosis assays can then be used to examine treated cells for evidence of apoptosis.
