Blotto, commonly made with non-fat dry milk, is a blocking solution widely used in various immunoassay techniques, including Western blotting and enzyme-linked immunosorbent assays (ELISA). Non-fat dry milk is dissolved in a buffer, often Tris-buffered saline (TBS) or phosphate-buffered saline (PBS), to create Blotto. This solution serves as a blocking agent to prevent non-specific binding of antibodies to the membrane or plate surface during immunoassays. Blotto effectively blocks unoccupied binding sites on the membrane or plate, reducing background noise and increasing the specificity and sensitivity of antibody detection. It is typically added after the primary antibody incubation step and before the secondary antibody incubation step in immunoassay protocols. In addition to non-fat dry milk, other blocking agents such as bovine serum albumin (BSA) or gelatin may also be used to prepare Blotto, depending on the specific requirements of the assay and the target antigen.
