Bcas2 encodes BCAS2, an evolutionarily conserved component of the Prp19-associated spliceosome complex that supports pre-mRNA splicing and splice site selection. BCAS2 also interfaces with transcriptional regulation and genome maintenance pathways, and has been linked to cell-cycle progression through modulation of checkpoint and DNA damage response signaling. In mouse systems, perturbation of BCAS2 can alter alternative splicing programs and downstream gene expression networks that influence proliferation, differentiation, and stress responses. Dysregulated BCAS2 expression or splicing defects involving BCAS2-associated complexes have been studied in the context of tumor biology and other disorders where aberrant RNA processing contributes to disease mechanisms.
BCAS2 CRISPR/Cas9 KO Plasmid (m) is a pool of plasmids designed for targeted disruption of the Bcas2 gene in mouse cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the Bcas2 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the Bcas2 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish BCAS2 protein expression.
This CRISPR knockout system enables efficient generation of Bcas2-deficient cell models for investigation of BCAS2 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
