Spot 14 HDR Plasmid (h) 는 2-3 plasmids를 포함하며, 매개의 plasmid는 homology-directed DNA repair (HDR) template 는 Spot 14 CRISPR/Cas9 KO Plasmid (h)로 인한 cut site와 대응됩니다.
매 HDR template은 두개의 800 bp homology arms이 디자인되여있어 Cas9-induced double strand DNA break site주변의 genomic DNA 와 결합합니다
매 HDR Plasmid 에는 puromycin resistance gene이 있어 stable knockout (KO) cells을 얻을수있으며 RFP (Red Fluorescent Protein) gene이 있어 형질주입효율을 관찰할수있습니다.
Puromycin resistance와 RFP genes는 두개의 LoxP sites 사이에 위치하여 Cre Vector sc-418923로 추가가공이 가능합니다.
