SB 743921 is a second generation, highly potent and active KSP inhibitor with a Ki of 0.1 nM for KSP (Eg5). SB 743921 is 5-fold more potent against the ATPase activity of KSP than ispinesib. SB 743921 has cytotoxic activity at less than 2 nM in a spectrum of tumor cell lines.
