PML 항체(PG-M3)는 웨스턴 블롯(WB), 면역침전(IP), 면역형광(IF), 면역조직화학(IHC)을 통해 인간 유래 PML 단백질을 검출하는 쥐 단일클론 IgG1 kappa 경쇄 항체입니다. PML(PG-M3) 단일 클론 항체는 비접합 형태와 아가로스, 호스래디시 퍼옥시다아제, 피코에리트린, 플루오레세인 이소티오시아네이트, 다중 알렉사 플루오르® 접합체를 포함한 다양한 접합체 형태로 제공됩니다. PML은 아연 핑거 전사 인자로서 기능하며, 세포 분화 및 세포 사멸과 관련된 유전자 발현을 조절하는 데 매우 중요합니다. 이러한 과정에서 PML의 역할은 특히 급성 전골수성 백혈병에서 중요합니다. 이 질환에서는 PML 유전자가 레티노산 수용체 α(RARα) 유전자와 염색체 전위를 겪으면서 PML/RARα 키메라 단백질이 형성됩니다. 이 비정상적인 단백질은 레티노산 의존성 분화에 필수적인 핵 공동 억제제-히스톤 탈아세틸화 복합체를 모집함으로써 정상적인 골수성 분화를 방해하고, 또한 유전자 과메틸화를 유도하여 중요한 유전자의 침묵을 유도하고 백혈병 발생을 촉진합니다. 이러한 경로에 영향을 미치는 PML의 능력은 정상적인 세포 기능과 질병 상태 모두에서 PML의 중요성을 강조하며, 따라서 PML(PG-M3) 항체는 이러한 메커니즘을 연구하는 연구자들에게 유용한 도구입니다.

Can I get an epitope sequence of sc966 anti-PML antibody? I would like to check if I could use it to blot mouse protein on IF. Thank you! Won

Thank you for your question. This antibody was raised against an epitope corresponding to amino acids 37-51 mapping near the N-terminus of PML of human origin. This antibody has not yet been tested in mouse and so that is not a recommended species at this time.

For molecular weight of PML, it displayed 55-90KD, others showed 90-132KD. So, what is the difference?

Thank you for your question. The PML protein has to main isoforms which run around 78 and 92kDa. Please contact your local distributor or technical service for further assistance.

Rated 5 out of 5 by from Very clear signal!We applied Anti-PML antibody (PG-M3) in immunofluorescence, to visualize PML bodies in U2OS cell line. Antibody at 1:1000 dilution in Goat serum blocking solution (1mg/ml BSA, 3% goat serum, 0,1% Triton X-100, 1mM EDTA ph 8.0) and O.N incubation produced a strong specific signal with no background. solution (1mg/ml BSA, 3% goat serum, 0,1% Triton X-100, 1mM EDTA ph 8.0).

Rated 5 out of 5 by from Good mAb for IFAAs noted in a lot of papers, this monoclonal antibody works well in IFA. Although This mAb has back-ground signal, the positive signal of PML is strong to obtain good picture. (HeLa cell, 1:100 dilution in 1% BSA/PBS, 4C overnight)
