PME-1 Antibody (E-8) is a mouse monoclonal IgG2a antibody that detects protein phosphatase methylesterase-1 (PME-1) in mouse, rat, and human samples through various applications including western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). PME-1 plays a crucial role in the regulation of protein phosphatase 2A (PP2A), a key enzyme involved in cell growth inhibition and cell cycle regulation, by catalyzing the demethylation and inactivation of this phosphatase. The ability of PME-1 to demethylate the catalytic subunit of PP2A is vital for maintaining proper cellular signaling and function, as dysregulation of PP2A activity is associated with various diseases, including cancer. PME-1 is conserved across species from yeast to humans and features a catalytic triad similar to that found in lipases, with an activated serine serving as the nucleophile at its active site. This structural similarity underscores PME-1′s importance in cellular processes and highlights PME-1′s potential as a therapeutic target. Additionally, the covalent modifications of carboxymethylation and demethylation that PME-1 mediates are influenced by electrostatic interactions at specific residues or metal ions near the active site, further emphasizing the complexity and significance of PME-1′s regulatory functions in eukaryotic cells.
