Neurabin-II Antibody (D-7) is a high quality monoclonal Neurabin-II antibody (also designated Neurabin-II antibody) suitable for the detection of the Neurabin-II protein of human origin. Neurabin-II Antibody (D-7) is available as both the non-conjugated anti-Neurabin-II antibody form, as well as multiple conjugated forms of anti-Neurabin-II antibody, including agarose, HRP, PE, FITC and multiple Alexa Fluor® conjugates. Neurabin-II, also called spinophilin, interacts with actin and PP-1 in dendritic spines of the central nervous system. The gene encoding human neurabin-II maps to chromosome 17q21-q22. The structural characteristics of neurabin-II include one F-actin binding domain at the N-terminal region, a predicted coiled-coil struture at the C-terminal, one PDZ domain at the middle region, and a domain known to interact with transmembrane proteins. Neurabin-II bundles actin fliaments in vitro. In vivo, spinophilin localizes to the cortical sites of actin filaments and to the sites of active membrane remodelling. Neurabin-II also forms a complex with the catalytic subunit of PP1 and modulates PP1 enzymatic activity in vitro. Neurabin-II localizes to the head of dendritic spines and aids in the ability of PP-1 to regulate the activity of a-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid (AMPA) and N-methyl-D-asparate (NMDA) receptors. In this manner, neurabin-II modulates both glutamatergic synaptic transmission and dendritic morphology. Synergistic interactions between spinophilin and human tumor supressor ARF suggest a role for neurabin-II in cell growth.

Why does neurabin II run at ~130kD, when the sequence predicts an 89kDa protein? Seems like a large difference.

Thank you for your question. The observed molecular weight is significantly higher than the predicted molecular weight. Please refer to the following publication as a reference for an approximately 140 kDa observed molecular weight for Neurabin-II, also known as Spinophilin, PMID: 9275233.

I am using Neurabin-II (D-7): sc-373974 monoclonal antibody to stain paraffin embedded tissue. Do you recommend antigen retrieval when using this antibody?

Great IF staining cortical neuronsStains cultured cortical neurons beautifully. Looks just like actin stain in the growth cone. First SPN antibody I have had that works.
These are 36hr cultures of mouse cortical neurons. (Yes, it works on mouse)
Use methanol fixation- (I do mine after PFA fixation) and the antibody at 1:100. Without methanol you can see a bit but its super faint.
