TMPO encodes lamina-associated polypeptide 2 (LAP2), an inner nuclear membrane protein that binds lamins and chromatin to organize the nuclear lamina and maintain nuclear envelope architecture. LAP2 isoforms contribute to tethering peripheral heterochromatin, regulating DNA replication timing, and coordinating post-mitotic nuclear reassembly, linking TMPO to cell-cycle control and genome stability pathways. Through interactions with lamin A/C and other envelope components, LAP2 influences mechanotransduction and nuclear integrity under proliferative or stress conditions. Altered TMPO/LAP2 expression or nuclear envelope dysfunction is associated with aberrant nuclear morphology observed in cancer and other disorders involving compromised genome organization.
LAP2 CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the TMPO gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the TMPO together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the TMPO open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish LAP2 protein expression.
This CRISPR knockout system enables efficient generation of TMPO-deficient cell models for investigation of LAP2 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
